Lymphocytes with or without Fc receptors (FcR⁺ or FcR⁻) from 14 healthy males were preincubated with or without lupus sera and then cultured for 7 days. Lymphocyte subpopulations were enumerated with monoclonal anti-Leu-7 (HNK-l) and OKT3 on day 1, day 4 and day 7. The study was designed to see: (1) Whether natural killer (NK) cells are generated from FcR⁻ lymphocytes? (2) Do NK cells shift to T cells during the 7- day- culture? and (3) The influence of lupus sera on the maturation of NK cells. Our results showed that no evidence indicated the generation of NK cell from FcR⁻ lymphocytes. There was also no evidence of shifting NK cells to T cells. Different subsets of NK cells, FcR⁺ & FcR⁻, with different ratio of functional immature NK cells were found. Preincubation with lupus sera did not cause the change of percentages of NK cells and functional immature NK cells during the culture period. It indicates lupus sera do not affect the course of NK maturation. NK cells with Leu- 7 (HNK-1) surface marker failed to show any significant change in a 7- day- cuI(ure. Concomittant study of lymphocyte subpopulations with anti- Leu- 7 (HNK-l), anti- Leu- 11 and OKT3 is needed to provide more informations in NK cell maturation and the relationship between NK cells and T cells. |
