柴胡(Bup1euri Radix)具解表、解熱、疏肝解鬱、升舉陽氣等效能。如與其他藥材配伍成小柴胡湯或大柴胡湯等漢方製劑則具有抗炎性,對難治之慢性肝炎、腎炎具相當高療效。主成分有 Saikosaponin a, b, c, d, Saikogenin E, F, G, α-Spinasterol, Stigmasterol, Δ7-Stigmastenol, Δ22-Stigmasterol, Adonitol。過去鑑別柴胡,除傳統的組織切片,粉末鏡檢及五官鑑別外,對其所含 Saponin 成分亦作典型的起泡試驗或 Liebermann 反應試驗,然含有 Saponin 的中藥材甚多,以上述方法實無法針對柴胡藥材作特異性的鑑別,更遑論應用於製劑以定性柴胡成分。本實驗以市售柴胡為對象,藉由薄層層析法的分離效果,嘗試分離出柴胡的特有成分及其圖譜特徵。為顧及藥材栽培地區、採收期不同及基原不易確定,五件實驗藥材分別由台灣地區北、中、南部之藥房購得,並探討其共同性。 |

In this experiment we developed three types of mobile phase systems in TLC to identify Bupleuri Radix and its Chinese preparations. The mobile phase systems consist of: Ethylacetate-Methanol-Water (10:2:1), n-Butanol-Glacial Acetic Acid-Water (4:1:2), Chloroform. In these TLC mobile phase systems, We can easily separate Bupleuri Radix in Long Dan Shieh Gao Tong(龍膽瀉肝湯), Jia Wey Shiau Yau Sann(加味逍遙散), Buu Jong Yih Chih Tong(補中益氣湯) . Conquently, we clearly identified the spots showing Bupleuri Radix by using P-Dimethlyamino benzaldehyde spray reagent or U.V. light (366nm). |
