An Assay of Angiotensin-Converting Enzyme Activity Obtained Using Capillary Electrophoresis

本研究在於開發一簡單的毛細管電泳方法，以測定ACE的活性。ACE反應採用hippuryl-L-histidyl-L-leucine（HHL）作為反應基質，以毛細管區帶電泳（CZE）分離及定量其產物hippuric acid（HA）。最適之CZE分離條件是在應用65 cm×75 mm I.D.熔矽毛細管（有效分離長度55 cm），以10 mM或20 mM磷酸鹽緩衝液（pH 6.0）為電解液、20 kV電壓及228 nm偵測波長。

The authors developed a simple method to measure angiotensin-converting enzyme (ACE) activity using capillary electrophoresis (CE). With hippuryl-L-histidyl-L-leucine (HHL) as the substrate, we separated and quantified the enzymatic product, hippuric acid (HA) using capillary zone electrophoresis (CZE). CZE was performed with a 65 cm fused-silica capillary (55 cm effective length, 75 μm I.D.), using a 10 or 20 mM phosphate solution (pH 6.0) as the running buffer with an applied voltage of 20 kV at room temperature. The HA was identified and monitored at 228 nm.
