The conditions of pretreatment for vitamine E analysis, especially the effect of the saponification of infant formulas with or without water added on the analytical result was studied. It was found that dl-x-Tocopherol acetate would not be completely transformed to dl-α-tocopherol if water was added to sample before saponification. High performance liquid chromatoqrophy with a U.V. detector was compared with that with a fluore scente detector for vitamine E detection. The results showed that fluorescence detector (EM 330 nm, EX 290 nm) had higher sensifivity forα-Tocopherol, while UV detector had higher sensitiving for its acetate form. Recovery analysis for three groups of foods were performed. Average recoveries forα-Tocopherol added to infant formalas, health foods and edible fats and oils were 100.4%, 98.5% and 97.0%, respectively. The lowest detection limit forα-Tocopherol by the above mentioned instrument was 0.2 ug/ ml. |
