乙二醇單甲基醚(ethylene glycol monomethyl ether,EGME )、乙二醇單乙 基酸(ethylene glycol monoethyl ether,EGEE )及其乙酸酯之毒性代謝物分別為甲氧基醋酸(methoxyacetic acid, MAA)、乙氧基醋酸(ethoxyacetic acid,EAA)。本研究是以混合溶劑萃取尿中MAA、EAA及內標準品丁氧基醋酸(butoxyacetic acid, BAA),再以氣相層析儀分析其甲基衍生物。MAA檢量線之線性範圍為5.0~200.0μg/ml,EAA為3.5~70.0μg/ml,且同日及異日檢量線準確度及精密度之變異係數均在9%以下;MAA的回收率為96.27%,EAA為101.69%;偵測極限MAA為0.776μg/ml,EAA為0.367μg/ml;當尿液檢體貯存於-20°C時,其穩定性長達五個月以上。 |
英文摘要 |
Methoxyacetic acid (MAA) and ethoxyacetic acid (EAA) are the major metabolites of ethylene glycol monomethyl ether and ethylene glycol monoethyl ether and their acetates, respectively. Samples were analyzed by gas chromatography after extraction from urine and then methylation. Butoxyacetic acid (BAA) was used as an internal standard. The linear range of MAA was 5 to 200 μg/ml and EAA was 3.5 to 70.0μg/ml. The recoveries of MAA and EAA from urine were 96.27 % and 101.69 %,respectively. The intra-assay and inter-assay variation were both less than 9 % . The detection limit was 0.776μg/ml for MAA and was 0.367μg/ml for EAA. The stability of the specimen was found to be at least stable for five months, when they were kept at -20°C. |
