Clonal propagation of Phalaenopsis and Doritaenopsis by internode of flower stalka in vitro were studied. 73.4% protocorm-like bodies (PLB) formed from epidermal cells, and/or parenchyma cells near the vascular bundles of internode tissue was found in the 1.2 times Vacin and Went's basal salts, with 1 mg/I BA. However, only 46.8% PLB formed when BA content increased to 5 mg/I. BA+NAA, decreased PLB formation. Kinetin+NAA or 2, 4-B, all thin slices of internodes blacked and necrotic. Subculture the PLB could get large number of PLB, and formed young plants thereafter. Young flower stalk with dividing epidermal cell and parenchyma cell near the vascular bundles are easy to form protocorm-like bodies in some species and hybrides of Phalaenopsis and Doritoenopsis. After the last floret anthesis, the mature internodes were not easy to form PLB in BA 1 mg/I medium. It seems likely, the longer time of flower stalks extend, the less possibility for cultured internodes will be, and induced the PLB formation percentage will decrease. After the last flower senescences, all internodes do not have ability of differentiation. |
