許多研究指出,草本植物為天然抗氧化物質的來源之ㄧ,而這些抗氧化物質在抑制突變及預防癌症的機轉可能具有相關性。金錢薄荷(Glechoma hederacea)為唇形花科植物,又名連錢草、活血丹、落地金錢等,為一傳統入藥與食用植物,民間傳用具有活血化瘀、解熱利尿等功效,但鮮少對其生理活性做探討。因此,本研究的主要目的是以體外試驗探討金錢薄荷熱水萃取物(hot water extract from G. hederacea, HWG)之抗氧化活性、致突變性以及抗致突變能力。金錢薄荷以熱水萃取3小時後冷凍乾燥。抗氧化活性測定項目包括α,α-diphenyl-β-picrylhydrazyl (DPPH) 、 2,2‘-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid)及超氧陰離子(super-oxide anion)自由基清除能力,亞鐵螯合能力,還原力以及抑制脂質氧化等效果。體外致突變性及抗致突變能力則以Ames test探討之。結果顯示, HWG具抗氧化特性,而且其對於清除超氧陰離子基及螯合亞鐵之能力甚至優於標準品VitC與Trolox。Ames test試驗結果亦顯示,HWG於測試濃度範圍內(0.31-5.00 mg/plate)對五種測試菌株(Salmonella typhimurium TA97, TA98, TA100, TA102以及TA1535)皆不具致突變性;此外,在添加S9酵素混和液的反應系統下,HWG則展現其抗致突變效力。HWG不僅具抗氧化活性,亦有抗致突變之潛能;然其活性成分仍待進一步分析。" |

Several studies have demonstrated that herbal plants contain various potential antioxidants and these antioxidant components may be potential antimutagens or anticarcinogens. Glechoma hederacea belongs to the Labiatae family. According to ancient Chinese prescrip-tions, G. hederacea was used for treating diuresis and stimulating blood circulation. However, there are few reports on its biophysical functions to date. The objective of this study was to evaluate the antioxidant, mutagenic and antimutagenic activities of the hot water extract of G. hederacea (HWG). G. hederacea was extracted with distilled water at 100°C for 3 h. The antioxidant activities tested included α,α-diphenyl-β-picrylhydrazyl (DPPH), 2,2‘-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) and superoxide anion radical-scavenging effects, Fe2+-chelating ability, reducing power and lipid peroxidation inhibition. The in vitro mutagenic and antimutagenic properties of the extracts were investigated using the Ames test. The results showed that HWG possessed antioxidant characteristics. Moreover, the anti-oxidant activities of HWG were significantly higher than those of Vitamin C and Trolox in terms of superoxide anion radical-scavenging activity and Fe2+-chelating ability (p < 0.05). The results of Ames test indicated that HWG had no mutagenicity toward all tester strains including Salmonella typhimurium TA97, TA98, TA100, TA102 and TA1535 with/without the metabolic activator (S9 mix). The extracts had marked inhibition effects against the mutagenicities of the diagnostic mutagens with the S9 mix system. The HWG showed antioxidant and anti-mutagenic potential. However, the active compounds of HWG need be further investigated. |
