Determination of 17α-methyltestosterone in Bovine Serum Using Liquid Chromatography Tandem Mass Spectrometry

本研究建立並確效高效液相層析串聯質譜儀（HPLC-MS/MS）方法，檢測牛血清檢體中之合成成長促進因子17α甲基睾丸酮（MT）。利用乙?（acetonitrile）單一步驟萃取（single-step extraction）後，藉由偵測其甲基睾丸酮之?衍生物，提升甲基睾丸酮測定能力。根據歐盟Decision No 2002/657/EC所訂準則，並以氘化睾丸酮（deuterated testosterone, T-d3）為內部標準品，本方法經確效之偵測極限值（decision limit）為0.05 ng/mL 、偵測能力（detection capability）為0.09 ng/mL 、重複性相對偏差（CV）為7.04% 、實驗再現性相對偏差（CV）為11.00%以及準確度為87.00%。"

A high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) based method has been developed and validated to confirm the presence of the synthetic growth promoter 17a-methyltestosterone (MT) in bovine serum samples. After a single-step extraction with acetonitrile, the enhanced determination of methyltestosterone was achieved by detecting its oxime derivative. In accordance with the criteria defined in Commission Decision 2002/657/EC and using deuterated testosterone (T-d3) as an internal standard, the method has been validated in terms of decision limit (0.05 ng/mL), detection capability (0.09 ng/mL), repeatability CV (7.04%), within-laboratory reproducibility CV (11.00%), and accuracy (87.00%).
