市售中毒草製劑溫清飲以30%甲醇於超音波60°C加熱振盪抽提後,利用高效液相層析法(HPLC),以去羥梔?(Geniposide)及芍藥?(Paeoniflorin)為指標成分,使用P-Hydroxybenzoic acid methyl ester為內部標準物質,於層析管RP-18( Lichrospher),移動相:乙?:水:醋酸 = 15:85:1,檢測器: UV(238nm, 0.1 AUFS),流速: 0.6ml/min之條件下,同時測定製造中梔子之Geniposide及芍藥之Paeoniflorin含量,結果得到良好的分析效果。回收率亦分別達97.5%與94.0%。本研究亦發現減壓濃縮乾燥式冷凍乾燥以及添加澱粉賦形劑等製造過程,對上述兩種成分之含量測定均未造成影響。上述分析條件,亦適用於加味逍遙散及防風通聖散等同時含梔子及芍藥之處方。" |
英文摘要 |
A high-performance liguid chromatographic method for Quantitative analysis of geniposide in Gardeniae Fructus and paeoniflorin in Paeoniae Radix was developed. The samples were separateel by a RP - 18 Lichrospher column eluted with water-acetonitrile-acetic acid (85:15:1, v/v)at a flow rate 0.6ml/min and were detected at UV 238nm. The method was applied to monitor the optimum conditions for the extraction of the traditional Chinese medicinal preparation 'Unseiin', which contains Gardeniae Fructus and Paeoniae Radix. In this study, methy parabebn was used as the internal standard. A good and reproducible separation of paeoniflorin and geniposide was achieved within 31 min. The method was also applicable to other preparations that contain these two medicinal plants such as Kamishoyosan and Bofutsuseisan. |
