In 1997, the discovery of Klotho led to identification as a protein involving longevity with broad therapeutic implication. Klotho, initially introduced as an anti-aging protein, but recent studies have suggested that Klotho is involv-ed in the progression of several types of human cancer. The soluble form of Klotho have diverse pharmacological functions such as anti-inflammatory, anti-oxidative, anti-fibrotic, tumor-suppressive etc. Soluble Klotho, through inhibition of the TGF-β, Wnt, FGF, IGF-1 signaling pathways, Klotho also inhibits tumorigenesis. However, further investigation is required to clarify detailed mechanisms of Klotho in cancers. There remains a long way to ad-minister the Klotho protein into clinical application. |
