The role of Klotho in the cancer

1997年發現Klotho乃一長壽蛋白，且有很多治療上用途。首先，Klotho被認為是一種抗老化蛋白，後來了解其在人類癌症上有關連性。溶解性Klotho有抗發炎、抗氧化、抗纖維化和抑制腫瘤等功用。可溶性Klotho經過抑制TGF-β，Wnt，FGF，IGF-1等信號途徑，有抗腫瘤效果，但仍需日後更大規模的研究，才能真正了解Klotho在癌症的各種用途上的功效。

In 1997, the discovery of Klotho led to identification as a protein involving longevity with broad therapeutic implication. Klotho, initially introduced as an anti-aging protein, but recent studies have suggested that Klotho is involv-ed in the progression of several types of human cancer. The soluble form of Klotho have diverse pharmacological functions such as anti-inflammatory, anti-oxidative, anti-fibrotic, tumor-suppressive etc. Soluble Klotho, through inhibition of the TGF-β, Wnt, FGF, IGF-1 signaling pathways, Klotho also inhibits tumorigenesis. However, further investigation is required to clarify detailed mechanisms of Klotho in cancers. There remains a long way to ad-minister the Klotho protein into clinical application.
