Metoclopramide及Bromopride之化學結構差異僅在於前者含氯原子,後者含溴原子;其紫外光吸收圖譜甚為相似;而薄層分析亦欠理想,至今二者間尚熟可靠之鑑別法可循。今利用逆相高效液相層析法,以Caffeine為內標準品,e-Bondapak Phenyl層析管固定相,乙睛-0.025M磷酸鹽緩衝液(20:80)為移動相,分離並鑑別、定量此二成分。本法之回收率Metoclopramide Bromopride分別可達98.5%及99.7%。為一簡便可靠之測定法。
Metoclopramide is a chlorinated analogue of bromopride, which is a bromo compound. The UV absorption spectrums of both compounds show great resemblance. Separation of them by thin layer chromatography is not yet worked out. Identification and determination of these two analogues in pharmaceutical preparations can be carried out satisfactorily by high performance liquid chromatography on a u-Bondapak Phenyl column with UV detector. A mixture of acetonitrile and 0.025M ammonium phosphate buffer was employed as the mobile phase. Caffeine was used as the internal standard. The method is rapid and reliable. |
