We used a rapid and reliable high-performance liquid chromatographic method for determination of tetramethylpyrazine in rat brain. The separation was achieved on a Cosmosil 5C18 column (4.6 x 250 mm, particle size 5 um). The mobile phase consisted of methanoi-water (50:50, v/v, pH 3.0 adjusted by orthophosphoric acid) at a flow rate of 1.0 mL/min. Tetramethylpyrazine was monitored at a wave- length of 280 nm. The lower limit of quantification in brain tissue was O.1 ug/g, and only a small amount of brain tissue (100 ug) was required for analysis. Concentrations of tetramethylpyrazine in various regions of the brain (cerebral cortex, brainstem, striatum, hippocampus, cerebellum and mid- brain) were not significantly different at 15 min afteradministration (10 mg/kg, i.v.). Mean concentra- tion of the tetramethylpyrazine in plasma was approximately five times greater than in brain tissue. In additional analysis, about 17% of tetramethylpyrazine may penetrate blood-brain-barrier. |
