An improved high-performance liquid chromatographic method was useded to determinate trilinolein, the active triacylglycerol principles of Ginseng Radix (Renshen), Acanthopanacis Radicis Cortex (Ciwujia), Angelicae Sinensis Radix (Danggui), Ligustici Wallichii Rhizoma (Chuanxiong), Bupleuri Radix (Chaihu), Paeoniae Radix Rubra (Chishao), Paeoniae Radix Alba (Baishao) and Salviae Miltiorrhiza Radix (Danshen). Chromatographic analysis is achieved on isocratic system consisting of a polymeric reversed-phase C18 column with a mobile phase of acetonitrile-methanol (50:50, v/v) to elute the trilinolein. The eluted trilinolein is detected at 205nm. The linear calibration range and detection limit for trilinolein were 0.5-20μg/mL and 0.1μg/mL, respectively. It was found that the trilinolein contained in n-hexane extracts of Ginseng Radix (Renshen), Acanthopanacis Radicis Cortex (Ciwujia), Ligustici Wallichii Rhizoma (Chuanxiong), Angelicae Sinensis Radix (Danggui), Bupleuri Radix (Chaihu), Paeoniae Radix Rubra (Chishao), Paeoniae Radix Alba (Baishao) were 0.274±0.009, 0.131±0.007, 4.135±0.071, 0.530±0.017, 0.168±0.005, 0.101±0.002 and 0.443±0.010mg/g, respectively. However, the trilinolein was not detected in the Salviae Miltiorrhiza Radix (Danshen). |
