Neoalsomitra integrifoliola has been used in Taiwanese folk medicine for the treatment of inflammation. In our previous works, we found that increased intraperitoneal vascu1ar permeability and paw edema were inhibited by ethyl acetate extract (EtOAc ext.) of N. integrifoliola. In this report, we further studied the effect of EtOAc ext. on PAF-induced biological phenomena. The following results were obtained: (1) EtOAc ext. depressed skin plasma extravasation induced by PAF, but not by bradykinin and compound 48/80. (2) Both PAF and arachidonic acid (AA) caused paw edema were inhibited by EtOAc ext. (3) EtOAc ext. inhibited pleurisy observed 6 hr , but not 30 min after PAF administration. (4) EtOAc ex t. inhibited anaphylactic shock caused by PAF in mice. (5) PAF-induced hypotension was not affected by EtOAc ext. (6) Platelet aggregation was investigated in rabbit in vitro. EtOAc ext. inhibited PAF-induced aggregation in dose-dependent manner, with an IC50 of 123 μg/ml. AT IC50, it completely inhibited both collagen-and AA-induced aggregation. Our findings suggested that the inhibition of EtOAc ext. on PAF might be due to inhibit the release of chemical mediators by PAF-stimu1ated cells. |
