Gomphocarpus physocarpus, commonly known as balloon plant or swan plant, is a perennial herbaceous ornamental crop of the subfamily Asclepiadoideae, family Apocynaceae. Gomphocarpus mosaic virus (GoMV), a new and emerging potyvirus, infects Gomphocapus spp. causing symptoms of mosaic, mottle and crinkling on leaves. Electron microscopic examination revealed the presence of filamentous virus-like particles about 750-850 x 12 nm in crude sap and typical potyviral pinwheel and laminated inclusion bodies in ultrathin sections of infected tissues. A GoMV isolate was mechanically inoculated to 24 plant species belonging to nine families to determine its host range. Fourteen species of plants out of 24 tested were affected. SDS-PAGE analyses revealed that GoMV contains a single viral coat protein subunit of a molecular weight about 30 kDa. Specific antiserum against the GoMV was produced by immunizing New Zealand white rabbit with purified virus particles. The specific antiserum reacted positively to GoMV in all tests using SDS-agar gel double diffusion assays, western blotting, and indirect enzyme-linked immunosorbent assay (ELISA). |
