"In this work, we combined traditional and modern techniques, i.e., morphological and species-specific DNA markers, respectively, to identify the codling moth, Cydia pomonella L. According to a published article, morphological characters, mainly the style of the proleg crochets and the number and distribution of setae, etc., were applied for identification of codling moth larvae. The rationale of sequence characterized amplified regions (SCARs) was adopted to develop DNA markers for identification of codling moths, and we obtained 5 pairs of species-specific PCR primers for this purpose. By applying these 2 methods, we successfully identified 2 codling moth specimens which were discovered in 2 batches of apples imported from the US in the course of quarantine inspections by the Kaohsiung Division of the Bureau of Animal and Plant Health Inspection and Quarantine (BAPHIQ) on November 6 and 12, 2002, respectively." |
