目的:本文旨在闡述TGF-β信號通路在主動脈剝離的發病中的機制,併與冠狀動脈疾病的指標比較。主動脈剝離致病病因至今仍不明朗。在主動脈剝離之發展中,TGF-β信號途徑的機制先前還沒有報道。
方法:本研究包括於本院接受外科手術治療之連續20例的主動脈剝離和20例的冠心病患者。其主動脈組織標本采集和TGF-β1、TGF-β受體以蛋白質印迹法檢測。
結果:蛋白質印迹法顯示TGF-β1、TGF-β受體均在兩組主動脈組織表現。蛋白質的表現水平在主動脈剝離比的冠心病患者主動脈組織高得多。定量相對灰度結果,TGF-β1、TGF-β受體同時在主動脈剝離的冠心病患者的表現是良好的正相關關系。
結論:主動脈剝離動脈瘤的發病機制是一個複雜的血管重塑過程與血管損傷和修復有關。主動脈剝離的血管重塑過程在TGF-β信號通路的性質、方式、機轉和強度方面可能與粥樣硬化性血管重塑有所不同。主動脈剝離的機制可能與TGF-β/Smad信號傳導途徑的功能的明顯增強有關。
英文摘要
Objectives: This article aims to demonstrate the underlying mechanisms of aortic dissection compared to those of coronary artery disease in terms of the TGF-β signaling pathway. The causative etiologies have remained uncertain until now. The mechanisms of the TGF-β signaling pathway in the development of aortic dissection have not been previously reported.
Methods: The subjects of the present study were 20 consecutive patients with aortic dissection and 20 patients with coronary artery disease undergoing surgical treatment in this hospital. The aortic tissue samples were collected and the TGF-β1 and its receptor TβRI were detected by Western blotting.
Results: Western blotting revealed the TGF-β1 and TβRI receptor were both positively expressed in the aortic tissue in both groups. The expressions of the proteins were much higher in the aortic tissue of aortic dissection than those of the coronary artery disease patients. The quantitative relative gray scales showed the expressions of TGF-β1 and TβRI in both the aortic dissection and coronary patients had good positive correlations.
Conclusions: The pathogenesis of aortic dissection is associated with a complex vascular remodeling process incorporating vascular injury and repair. The aortic remodeling might differ between aortic dissection and atherosclerosis in terms of the nature, mechanism, mode and intensity of the TGF pathway activities. The mechanisms of aortic dissection might be related to the significant enhancement of the functions of the TGF-β/Smad signaling pathway.
