It is now known that Tristeza is the only virus disease transmitted by aphid vector and the control of this virus disease by using virus free budwood becomes a critical factor .in many areas where the citrus have got different mixtures of virus. The author has proved from several tests that the disease in Taiwan groves is a mixed type, so we need a rapid method determine the virus free citrus trees for controlling Tristeza in this The purpose of this test is to develop a rapid Colorimeter and Snectronhoto meter method for assaying the virus content which is based on the polyphenol and nucleic acid incrased in the tissue of virus infected trees. The results this test are summarized as follows: 1. The Photoelectric colorimeter is available for determining the polyphenol content of Ponkan leaf-tissue. It has been found in this test that the filter Klett No. 42 has more conspicuous readings than the filter Klett No. 54. The range of polyphenol content shown by the leaf-tissue of virus free seedlings is 90.8±1.9Smu., and the range shown by the artificial inoculation trees is 200.6±20.43mu. 2. The xylem-tissue test with Photoelectric Colorimeter (filter Klett No.42.) showed higher range of readings than the leaf-tissue test. There are 143.9±3.74mu in virus free seedlings, and 316.S±3,74mu in artificial inoculation trees. 3. The polyphenol test with Photoelectric Spectrophotometer in visible range showed maximum absorption differences between the virus free seedlings and virus inoculated trees. The virus free seedlings occurred in range of 440~500mu; and the virus artificial inoculation trees occurred in the range of 460~510mu. The xylem-tissue test with Photoelectric Spectrophotometer got the same result. 4. The Nucleic acid test with the Photoelectric Spectrophotometer on leaf-tissue of Ponkan, the maximum absorption of virus free seedlings occurred in the range of 450~500mu., and the virus artifical inoculation trees occurred in the range of 470~520mu. |
