The effects of medium composition on seed germination and seedling development in vitro were studied in Phaius tankervilliae when seeds were harvested 130 days after pollination. The morphology and anatomy of seeds and germination process were also observed under light and scanning micrograph. Seeds sterilized with 0.5% NaOCl solution for 15 minutes could enhance germination percentage up to 63.6%. Tryptone 2g/l could promote and eliminate protocorm browning during embryo development under 1/4 MS, VW, and Hyponex (7N -2. 6P-15.8K) 2g/l basal salts with 2% sucrose. Medium without calcium (such as Thomale's GD1) and enriched with various kinds organic additives except 50ml/l coconut water would increase protocorm browning in embryogenesis of Phaius tankervilliae. Growth of seedlings were inhibited by benzyl adenine, adenine, citric acid and activated charcoal. Root formation and root growth were strongly inhibited in medium containing activated charcoal. It seems likely that activated charcoal strongly influence the balance of plant growth regulators in medium. |
