Bulb scales of Lycoris au rea Herb. were cultured year-round in ordor to find the best time for excision. Scales cultured in May gave the highest percentage of explants producing adventitious buds and these cultured in August and September the least, and the highest percentage of browning of scale explants. Murashige and Skoog medium (1962) supplemented with 2 or 3 mg/1 BA and 0.5 mg/1 NAA gave the highest percentage of survival under day-light fluorescent tubes with light intensely of 2000 lux under a light-dark cycles of 16/8 hours at 27 ± 2°C. Adventitious buds appeared in the axile of the twin-scale explant after 2 months of culture. Transferring the scale explant with adventitious bud s t o medium without any growth regulator and incubation at 20 ± 1°C promoted the development of adventitious buds into plantlets with a bulblet due to the thickening of leaf base. From the twin-scale ex plants excised from a mature bulb of 5 cm diameter and 8 cm height, 50-75 plantlets could be regenerated. Regeneration of more than 100 plantlets from a mature bulb is possible provide the excision is done in May and decapitation of regenerated bulblets was used to hasten the formation of adventitious buds. |
