香蕉RNA抽取方法之探討

Improved Method for the Isolation of RNA from Banana

香蕉是一種富含多醣類及酚類化合物之果實，故在抽取RNA的過程中會受到嚴重的干擾，致使RNA非常容易降解或是品質不佳。本研究嘗試SDS、SDS-LiCl、guanidine及CTAB（cetyltrimethylammonium bromide）等方法抽取香蕉之RNA，發現CTAB法最快速方便，由電泳分析及北方雜交分析顯示CTAB法所得之RNA品質優良，可進一步用以構築cDNA庫。

The isolation of intact RNA from the banana fruit tissue is extremely diffcult due to the high content of polysaccharides, polyphenolics and RNase. Evaluation on different isolation protocols with the application of SDS, SDS with LiCl, guanidine and CTAB respectively was carried out for the preparation of RNA from different organs of banana. We demonstrated by electrophoresis and northern analysis that the CTAB method is the most rapid and reliable one and can give high yield and good quality of RNA. By this method and oligo-dT cellulose chromatography, the polyadenylated RNA from ripening banana fruit pericarp has been successfully purified and used for the construction of cDNA library.
