The isolation of intact RNA from the banana fruit tissue is extremely diffcult due to the high content of polysaccharides, polyphenolics and RNase. Evaluation on different isolation protocols with the application of SDS, SDS with LiCl, guanidine and CTAB respectively was carried out for the preparation of RNA from different organs of banana. We demonstrated by electrophoresis and northern analysis that the CTAB method is the most rapid and reliable one and can give high yield and good quality of RNA. By this method and oligo-dT cellulose chromatography, the polyadenylated RNA from ripening banana fruit pericarp has been successfully purified and used for the construction of cDNA library. |
