Protocorm-like bodies (PLBs) of Doritaenopsis were cultivated in various types of bioreactors including air-driven periodic immersion (API) bioreactor, rolling bottle, spinner flask and airlift bioreactor. The best results were given by API bioreactor and rolling bottle among all tested bioreactors. Design and composition of inner container used in API bioreactor also affected proliferation and differentiation rate of PLBs. The proliferation rates of four different cultivars propagated in API bioreactor were ranged from 4.2-6.5 folds /month by weight. After one harvest, each API container could generate over 100 g of PLBs, which was estimated 3000-5000 plantlets at least. Our results indicate the increase of biomass from rolling bottle system is similar to the API bioreactor as well. Therefore, rolling bottle can be used for mass micropropagation of PLBs on Doritaenopsis or Phalaenopsis as another choice beside API bioreactor. |
