本研究試以花寶基本培養基為配方添加活性碳對蝴蝶蘭(Phalaenopsis)子瓶苗生長有促進之作用。三品種蝴蝶蘭瓶苗定瓶後,培養於四種光度(20、40、60及80μmol m-2 s-1 photosynthetic photon flux,PPF)下,葉輻以20μmol m-2 s-1 PPF處理者明顯較大,但其Fv/Fm值明顯較低,因此建議瓶苗馴化光強度為40-80μmol m-2 s-1 PPF。蝴蝶蘭出瓶苗於20℃下黑暗貯運3-12天,貯運3日失重率為1.7-2.3%,貯運12日則達5.4-7.1%。經種植十週後之存活率均達96.4%以上。而葉輻以Dtps. I-Hsin New Girl‘KH5250’及Dtps. Leopard Prince‘KH5733#2’經貯運9及12日處理者顯著較小。以10、15、20、25或30℃貯運6日,Dtps. Leopard Prince‘KH5733#2’及Phal. Join Angle‘TH274’以30℃貯運之失重率顯著較高(4.7及6.1%),Dtps. Sinica Sunday‘SCL141’以25及30℃貯運之失重率顯著較高(5.9及6.6%)。貯運結束後,以10℃處理者對三品種蝴蝶蘭有寒害發生。三品種蝴蝶蘭以10℃貯運者經種植10週之存活率明顯較低(78.0-92.4%),且生長較緩慢。建議蝴蝶蘭出瓶苗之外銷貯運溫度以15-20℃為最佳。
The Hyponex basal medium with addition of activated charcoal improved the growth of in vitro Phalaenopsis plantlets. In vitro Phalaenopsis plantlets were subcultured under one of four light intensities for acclimatization. Plantlets grown under 20μmol m-2 s-1 PPF had the widest leaf span, but lowest fresh weight and Fv/Fm value than those grown under 40, 60 or 80μmol m-2 s-1 PPF. The fresh weight of deflasked in vitro plantlets in three Phalaenopsis clones were measured after being stored at 20℃ for 3-12 days. The weight loss was 1.7-2.3% after 3 days and 5.4-7.1% after 12 days. Regardless of the temperature, survival rate reached 96.4% after 10 weeks of growing in a greenhouse. However, the leaf span was smaller in Dtps. I-Hsin New Girl ‘KH5250’ and Dtps. Leopard Prince ‘KH5733#2’ after stored for 9 or 12 days prior to planting. Deflasked in vitro plantlets of three phalaenopsis were stored at 10, 15, 20, 25 or 30℃ for 6 days. The weight loss of plantlets was the highest (4.7-6.6%) when stored at 30℃. After storage, all three clones stored at 10℃ showed symptoms of chilling injury. However, only Phal. Taisuco Kaaladian ‘186’ stored at 15℃ exhibited chilling injury. Plantlets stored at 10℃ had low survival rate (78.0-92.4%) and slow growth rate after 10 weeks of planting. Temperature ranging from 15-20℃ for exporting in vitro phalaenopsis plantlets is recommended. |
