本實驗以星辰花組織培養繁殖的芽體為試驗材料,探討不同培養基組成與型式、透氣度各異的培養盒,對試管苗玻璃質化與增殖率造成的影響。結果顯示,含有洋菜的培養基,不論培養盒的型式,都比含有水晶洋菜的培養基,在相同處理下,可以有效降低玻璃質化培植體的發生,提高芽體增殖率。L型培養盒及封石蠟膜的GA-7培養盒有較高的玻璃質化比率和較低的增殖率,可能與容器透氣性有關。而D型與未封石蠟膜的GA-7培養盒具有相似的培養效果,較適宜作為星辰花的培養盒。C8(BA 1.1 mg/l,NAA 0.5 mg/l)培養基比C1(IAA 4.0 mg/l,kinetin 2.5 mg/l)培養基有較高的增殖率,但亦有較高的玻璃質化植株比例。
The aim of this study is to investigate the combined effects of medium composition, size, shape and ventilation of various vessels on the degree of vitrification and proliferation of tissue-cultured statice. The results indicated that vitrification was effectively reduced and proliferation rate was improved on agar-solidified medium than medium with gelrite. Explants in GA-7 vessels with parafilm and L-type vessels had higher vitrification frequency and lower proliferation rate than other treatments. It may be related to poor ventilation of culture container. Treatments in D-type and GA-7 vessel without parafilm showed similar results, which is suitable for statice micropropagation. More shoots were regenerated on C8 (BA 1.1 mg/l, NAA 0.5 mg/l) than on C1 (IAA 4.0 mg/l, kinetin 2.5 mg/l) medium, however, less vitrified explants were produced on C1 than C8. |
