本研究以三個彩色海芋品種繼代培養之培殖體及發根苗為材料,調查連續繼代過程組培體增殖倍率並探討繼代次數對增殖族群主芽基部及發根苗基徑大小頻度分佈的影響。試驗結果顯示,彩色海芋莖頂組織培養芽體之增殖係由側芽直接發育而來,連續12次繼代過程中其增殖倍率隨繼代次數增加的變化,其中品種'Pacific Pink'與'Black Magic'表現呈同樣的趨勢而異於'Extra Gold',顯示在相同的培養條件下增殖倍率隨不同繼代次數的變化有品種間的差異。增殖族群主芽基部大小隨繼代次數的增加而下降,但參試的3個品種間所呈現的下降差異群發生在不同的繼代次數且主芽基部大小的頻度分佈隨繼代次數增加有愈趨集中的趨勢。發根培養階段對'Extra Gold'及'Pacific Pink'除了促進其發根、長葉外亦可增加其苗基徑的生長,且這種能力在12次的繼代中'Pacific Pink'苗基徑的生長增加是隨繼代次數的增加而加大,'Extra Gold'則在第9次繼代開始有較顯著的增加,'Black Magic'則發根苗與增殖苗基徑平均值幾乎相同。
Shoot apices of calla lily (Zantedeschia) were used for in vitro culture. Multiplication rate on lateral bud formation was low. There were two patterns of multiplication rates with serial subcultures among three calla lily cultivars. Among cultivars, cuttings' basal size declined with increased number of serial subcultures in vitro and showed significant differences at different subculture generations in 12 serial subcultures. The relative frequency of cuttings was concentrated with increasing number of serial subcultures in vitro. Rooting culture stage in vitro showed more effects on enhancing plantlets' basal size in 'Pacific Pink' and 'Extra Gold' than in 'Black Magic'. Plantlets' basal size increased within the increased number of serial subcultures in 'Pacific Pink'. In 'Extra Gold', the increment was after the 9th serial subcultures. There was no difference between plantlets' basal size and increased number of serial subcultures in 'Black Magic'. |
