Simultaneous Determination of Morphine HCl, Ketamine HCl and Droperidol in 0.9% Sodium Chloride by HPLC

於強酸、鹼及過氧化條件下，以高壓液相層析之安定性指示分析法，可同時定量0.9%氯化鈉水溶液中之morphine HCl 、ketamine HCl 及droperidol。本法採用BDS C18層析管柱（3.9mm×15 cm），移動相之組成為甲醇／乙A/0.01M 1-octane sulfonic acid sodium於0.005 M 磷酸緩衝溶液（pH4.0）（50/50/100），紫外線偵測器設定於254 nm，具良好之選擇性及專一性。

A simple and rapid liquid chromatographic method is presented for the determination of morphine HCl, ketamine HCl and droperidol in 0.9% sodium chloride injection stored in polyvinyl chloride (PVC) infusion bags. Assay was performed on a pre-packed HPLC Hypersil BDS C18 3.9 mm × 15 cm, particle size 3 μm column under controlled ambient temperature. Separation among morphine HCl, ketamine HCl, droperidol and their associated degradation compounds was achieved by isocratic elution using mobile phase consisting of methanol/acetonitrile/0.01 M 1-octane sulfonic acid sodium in 0.005 M phosphate buffer (pH 4.0) (50/50/100) at a flow rate of 1.0 ml/min. An UV/VIS variable programmable wavelength detector set at 254 nm was used. Sample volumes of 20 μL were injected. There was no need for sample pre-treatment.
