A high performance liquid chromatographic (HPLC) method was developed for the simultaneous determination of the steroid saponins, furostanol and spirostanol glycosides in Taiwanese yam cultivars. C18 column kept at 45ûC was used as the stationary phase. The mobile phase was a binary solvent system consisting of methanol and deionized water (62/38, v/v) in the first 20 min and 71/29 (v/v) from 21 to 65 min at a flow rate of 1 mL/min. Both evaporative light scattering detector (ELSD) and UV (203 to 215 nm) were employed and compared for the detection of saponins. The detection limits for ELSD and 203 nm UV were 2~3.5 mg and 0.1~0.2 µg, respectively at signal-to-noise ratio of 10. The recoveries of furostanol and spirostanol glycosides in the range of 0.25~3 µg were all above 92%. The presence of steroid saponins in Taiwanese yams was also determined.
本研究建立一套可同步測定台灣產山藥中之furostanol和spirostanol glycosides固醇類皂素之高效能液相層析法。實驗是以C18管柱為靜相,管柱溫度為45°C,以梯度溶劑為動相,甲醇/水= 62/38 (0-20分鐘),71/29 (21~65分鐘),並配合蒸發激光檢測器或紫外光偵檢器(203-215 nm)檢測,可得良好的分析結果。於信號值比雜訊值(S/N)等於10的條件下,以蒸發激光檢測器和203 nm紫外光檢測其偵測極限分別為2-3.5 µg和0.1-0.2 µg,furostanol和spirostanol glycosides的回收率皆高於92%。本研究亦利用所建立之方法檢測了台灣產山藥中固醇類皂素的含量。 |
