Baclofen activates metabotropic GABAB receptor, a G-protein coupled receptor, interacts with Ca2+ and K+ channels by second messenger systems, are resistant to the drugs modulating GABAA receptor. The overall inhibitory effect is reached by decreasing the likelihood of the action potential on central neurons [1]. Baclofen is rapidly absorbed through the digestive tract, reaching peak blood concentration in two hours after ingestion. Only 10%-15% of baclofen is metabolized in the liver, while the remaining 85%-90% is excreted unchanged by the kidneys. |
