Objective: This study tried to explore the association of single nucleotide polymorphisms (SNP) rs886205, rs671, and rs2106697 in aldehyde dehydrogenase 2 (ALDH2) gene with gout disease.
Methods: 1128 gout cases and 1769 matching controls from male Han Chinese were chosen to achieve a case-control study. SNPs of ALDH2 gene were genotyped using polymerase chain reaction-ligase detection reaction (PCR-LDR) analysis. Clinical, biochemical data and alcohol consumption were recorded to analyze the associations with ALDH2 genotypes and allele frequency.
Results: (1) Genotype frequency of the three SNPs in ALDH2 gene did not deviate from the Hardy-Weinberg equilibrium. (2) The allele frequency distribution of rs886205 was significantly different between gout patients and controls (p=0.031); the G allele of rs886205 appeared to be the risk allele for susceptibility to gout (OR=1.20, 95% CI=1.02-1.41), but the genotypes did not show the same significant result (p=0.094). Furthermore, the genotype and allele frequency distributions of rs671 show significant differences between gout patients and control group (both p<0.001). The G allele of rs671 appeared to be the risk allele (p<0.001, OR=1.66, 95% CI=1.41-1.96). (3) The mean blood sugar value was significantly different between individuals carrying different genotypes of rs671 in gout patients (p=0.024).
Conclusion: The polymorphisms rs886205 and rs671 are associated with susceptibility to gout in Chinese Han males, and rs671 may be related to the mean level of blood sugar of gout patients.
