Matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS) is an accurate, and cost-effective method used for identifying microorga nisms but its application was limited for identifying Helicobacter pylori (HP) for insufficient databases. In this study, we used previously sequenced 100 clinical HP isolates to generate mass spectra using MALDI-TOF M S. We used Flex Analysis and MALDI Biotyper 3 to plot phylogenetic trees and found 27 different types in ad dition to the 7 types in the Bruker database. These 27 types were added to the existing database, which resulte d in a significant increase in identification scores. We further tested the updated database using 20 other clini cal HP isolates, and the identification results were highly accurate (95.0%). Our establishment of the c omprehensive database significantly improved the accuracy of identifying HP using MALDI-TOF MS in the future. |
