"Stem rot disease of sweet pepper was found in Nantou and Yunlin counties in 2010. Water-soaked, longitudinal, and brown lesions were observed on the surface of stem, and the pith showed necrotic and disintegrated symptoms. Bacteria isolated from the infected tissues were verified to cause stem rot of sweet pepper in pathogenicity test. The pathogen was further identified as Pectobacterium carotovorum subsp. carotovorum (Pcc) based on physiological and chemical tests, Biolog identification, and 16S-23S rDNA intergenic spacer region sequences. Polymerase chain reaction using the primer pair Ec3F/Ec4R was developed for rapid diagnosis of Pcc. The 497-bp DNA fragment corresponding to Pcc can be amplified from the mechanically inoculated sweet pepper plants. The bactericide oxolinic acid has the most efficiency to inhibit Pcc growth." |
