滇藏方枝柏萃取液對抗大腸直腸癌細胞之抑癌作用機轉

The Anti-Cancer Effects of Juniperus indica bertol Extract in Colorectal Cancer

根據衛生福利部民國 108 年公佈之統計資料，大腸直腸癌 在國人十大癌症死因排名第三名，造成國人嚴重 的健康威脅及社會成本。近期文獻證實刺柏屬 （Juniper us） 植物具有保肝、抗氧化、抗發炎、抗菌及抗癌 等生物活性，並在肺癌、乳癌、肝癌及大腸癌細胞 都具有抑制生長之效果，但滇藏方枝柏 （Juniperus indicabertol, JIB） 至今尚未有完整的抑制大腸直腸癌的相 關研究，因此本實驗目的為探討 JIB 萃取液對抗 大腸直腸癌細胞之抑癌作用機轉。結果顯示 JIB 萃取物能夠有效抑制大腸直腸癌細胞生長，其主要透過調 控細胞週期相關蛋白（p53/p21 與 CDK4/Cyclin D1）及外在、內在細胞凋亡相關蛋白 （Caspase-8, 9, 3） 的表 達，誘發細胞週期停滯於 G0/G1期以及細胞凋亡，進而抑 制大腸直腸癌細胞生長。本研究實驗結果提供天然 JIB 萃取液抑制大腸直腸癌細胞機制探討外，同時顯示 JIB 萃取液具有潛力用於大腸直腸癌的預防。

According to the statistics of the Ministry of Health and Welfare, colorectal cancer (CRC) is the third leading cause of cancer-related deaths in Taiwan, causing serious health problems and social costs. Recent studies confirm that Juniperus species show biological activities o f hepatoprotection, anti-oxidant, anti-inflammation, anti-bacterial and growth inhibition effects in lung cancer , breast cancer, liver cancer, and colorectal cancer cells. Juniperus indica bertol (JIB) is one of Juniperus species, and the anti-cancer and mechanism of JIB extract in CRC has yet to be clarified. Therefore, the purpose of this study is to explore the anti-cancer effects of JIB extract against CRC cells. The results showed that JIB extract effectively inhibited the growth of CRC cells, induced cell cycle arrest at G0/G1 phase by regulati ng p53/p21 and CDK4/Cyclin D1 proteins expression and induced apoptosis by extrinsic and intrinsic apopto tic pathways. The results of this study provide a probing on the mechanism of natural JIB extract to inhibit CRC cells, and also indicate that JIB extract has high potential for prevention of colorectal cancer.
