Purpose: Hyperuricemia is considered to be the biochemical basis of gout. Urate oxidase, or uricase, is a new branch of drugs used for the treatment of hyperuricemia. Here we review several recently published evidences on the safety and efficacy of these drugs.
Findings: Rasburicase and pegloticase are the main uricosic drugs considered to be more effective than traditional urate lowering drugs. The main adverse reaction of uricase is immunogenicity. Other common adverse reactions are fever, nausea, headache, vomiting, rash and cardiovascular events.
Conclusion: Uricase is more effective at lowering urate. However, a significant reduction in immunogenicity is a key objective in the development of new uricases.
