Three cultivars of Gladiolus, Spic and Span, White Friendship, and Traveler were used for this experiment. Shoot tips of the vegetative lateral buds were excised from flower stalks approximately six weeks after the first floret opened. The explants of 300 to 500μ in height were cultured on Knudson's (1946) or diluted Murashige and Skoog's (1962) medium in which the explants developed into plantlets within 6 to 8 weeks. Those cultured on the media supplemented with 0.4 to 0.8 mg/e of 6-benzyladenine (BA) neoformed cormel-like bodies. These bodies could be divided to accelerate multiplication. Stock tissue used for propagation was stored on the MS medium supplemented with 0.8 mg/e of BA and subdivision series proceeded in every 3 to 4 weeks. Leaves, root and corms differentiated gradually from stock tissue when transferred to the medium without BA. The possibility of horticultural applications through vegetative shoot tip culture of Gladiolus has been discussed. |
