For improving the low seedling rates but high quality in Kaoshiung 1, and high seedling rate but low quality in red-petiole taro varieties, the technologies of taro protoplast isolation and fusion were developed to improve the production of taro. Maximum number of protoplasts was obtained when taro leaves or petioles were treated with enzyme solution containing 10% of mannitol, 0.2% of CaCl₂‧ 2H₂O, 1% cellulase and 0.5% of macerozyme for 10 hours. Further purification of protoplast could be achieved in 15~17% of sucrose solution. Best fusion inducer was found when 50% of PEG 6000 was added to 7mM of CaCl₂•2H₂O. |
