This study program was supported by a grant from the Council of Agriculture, Executive Yuan, Republic of China. The author went to Florist and Nursery Plants Research Unit, Beltsville Agricultural Research Center, United States Department of Agriculture to study gene transformation techniques from June to September, 1993. During this period, woody ornamental plants and chrysanthemum were used as testing materials, The program included DNA isolation, gene transformation via electrophoresis, electroporation, particle gun; and histochemical staining for GUS activity assay. This report describes various methods of gene transformation used in Florist and Nursery Plants Research Unit, BARC-USDA. |
