Effect of plant growth regulators on initiation of purple calli from tuber, stem, node, and leaf explants of Dioscorea alata L. var. purpurea M. Pouch cultured on MS (Murashige and Skoog, 1962) media were studied. Adding 2,4- D and kinetin to the MS media promoted more purple calli than that of NAA and BAP. The best results were observed among treatments containing high concentrations of 2,4-D and low concentrations of kinetin. Supplements of 10 μ M 2,4-D and 0.1 μ M kinetin to the MS medium induced more purple calli than those of other combinations. Tuber and node explants initiated more purple calli than stem or leaf explants. Purple calli subcultured on the liquid medium enhanced the production of purple cells. However, purple calli subcultured on semi-solid medium resulted in browning. |
