The growth and establishment of new lateral shoots of miniature roses (Rosa chinensis Jacq. minima, cvs., Red Sunblaze and Royal Sunblaze), from nodal explants was enhanced by the presence of parental stem tissue. Fresh and dry weight of shoots from a vertical, inclined or horizontal explant were greater than that those from an inverted explant. Fresh and dry weight of shoots and rate of lateral bud sprouting from explants from the upper five nodal segments were greater the lower two nodal segments. Under 5 to 40 μmol m-2s-1 lighting condition, the higher the photosynthetic photon flux (PPF) was the more quickly explants grew, matured and senesced. Initial growth was favored at a PPF of 20 μmol m-2 S-1. Twenty five combinations of 6-benzyladanine (BA) and 1-naphthale- neacetic acid (NAA) concentrations were tested for eight cultivars using selected explant and PPF conditions. Choice of medium for explant growth was dependent on cultivar: optimal growth of 'Red Sunblaze', 'Baby Katie', 'Central Gold', 'Green Ice', 'Orange Sunblaze', and 'Sequoia Gold' were achieved on a medium contain ing NAA at 0-0.1 mg 1-1 plus BA at 0.1 mg 1-1. However, the best growth response of 'Lavender Jewel' and 1 Royal Sunblaze' wasa achieved with NAA at 0-0.1 mg l-1 plus BA at 1.0 mg 1-1. Shoot proliferatoin was maximied when 9-16 explants were placed in each vessel. |
