The combined usage of autoradiography and the histological processing of diffusible substances was adopted in studying the distribution in detail of some labelled nutrients within a particular plant organ or in a portion of a plant or a fruit and the metabolic pathways in the whole plant. The techniques can also be used for studying the residual chemicals for plant protection as well as the accumulative status of some toxic metals in plant body due to contamination. Strawberry and tomato fruits, stem of Vitis venifera and cap of Pleurotus Florida were used as the materials and ³⁵S, ⁹⁰Sr, ¹⁴C and ²⁰³Hg were used as the tracing radionuclides in this research. After absorption, the histological processing of the samples was carried out by frozen sectioning and sublimate-dried. The autoradiography was achieved by dry mounted onto the Fuji ET-2F nuclear emulsions through apposition or wrapping method. The autoradiograms induced by the radionuclides displaying in various tissues in cellular level indicated the detail true. Results obtained in this study also revealed the acceptable histological quality and resolute autoradiograms of some radionuclides which may confirm the feasibility of the techniques in solving the problems mentioned above. |
