The study on shoot meristem culture of Allium sativum L. was conducted. The preparation of the fall strength culturing medium was as the following: Ca (NO₃)₂ 510mg; KC1 240mg; KH₂PO₄ 120mg; MgSO₄ 7H₂O 720mg; NH₄NO₃ 180mg; Na₂-EDTA 37.3mg; FeSO₄ 7H₂O 27.8mg; micro elements of Murashige & Skoog's medium; adenine 5mg; biotin O.Olmg; Ca-pantothenate 10mg; cystin 10mg; casein hydrolysate lmg; myoinositol O.lmg; nicotinic acid lmg; pyridoxin lmg; thiamin lmg; and glucose 30gm in one liter. The 100 to 200μ height of garlic shoot apical meristems were initially cultured in 1/3 strength of the culturing medium, under 25°C, and 0 to 150 lux light intensities for one month, and then were transferred to the full strength of the medium containing 0.1 to 1 ppm of NAA; under 25°C and 500 to 2200 lux light intensities. The plantlets obtained were strong and no virus symptomes. |
