本試驗以'Shine'長壽花的花蕾為培植體,利用體外培養之方式,探討花蕾長度與6-芐氨基腺嘌呤(BA)、萘乙酸(NAA)濃度對誘導芽體形成之影響。長壽花四種不同花蕾長度(3.0-4.9、5.0-6.9、7.0-8.9、9.0-10.9 mm)培養在同時含有BA及NAA之1/2 MS培養基下,皆可以再生芽體。使用3.0-4.9 mm及5.0-6.9 mm之花蕾培養在相同NAA濃度下,4 mg.L^(-1)BA相較於0-2 mg.L^(-1)BA處理,可以得到較高的芽體形成率,並且以越幼齡花蕾作為培植體,BA與NAA比值需要提高才能獲得較佳的芽體形成率。長壽花9.0-10.9 mm之花蕾培植體,以1 mg.L^(-1)BA及0.5 mg.L^(-1)NAA進行培養,所獲得的平均芽體數為最高;而7.0-8.9 mm之花蕾則以2 mg.L^(-1)BA + 0.5 mg.L^(-1)NAA及1 mg.L^(-1)BA + 0.5 mg.L^(-1)NAA處理為最佳;當花蕾長度減少為3.0-4.9 mm時,則以1 mg.L^(-1)BA+ 1 mg.L^(-1)NAA及4 mg.L^(-1)BA+ 1 mg.L^(-1)NAA之處理為佳。此報告可做為後續基因轉殖或誘變育種初代培養之參考。
The aims of this study is to investigate the flower bud length, 6-Benzylaminopurine (BA) and α-naphthaleneacetic acid (NAA) concentration in vitro on the shoot induction of Kalanchoe blossfeldiana 'Shine' using flower buds as explants. The shoots were induced from flower buds (3.0-4.9, 5.0-6.9, 7.0-8.9, and 9.0-10.9 mm in length) cultured on the half of MS medium supplemented with BA and NAA. More shoots induction was obtained in 3.0-4.9 mm and 5.0-6.9 mm flower buds cultured with 4 mg.L^(-1) BA compared with 0-2 mg.L^(-1) BA treatment in same NAA concentration. The higher BA to NAA ratio resulted in increased percentage of shoot induction when using young flower buds as explants. BA at 1 mg.L^(-1) combined with 0.5 mg.L^(-1) NAA resulted in most shoot number per explant in 9.0-10.9 mm flower buds as explants. The medium containing 2 mg.L^(-1) BA + 0.5 mg.L^(-1) NAA or 1 mg.L^(-1) BA + 0.5 mg.L^(-1) NAA yielded most shoot number per explant in 7.0-8.9 mm flower buds as explants. In contrast, most shoots were obtained in 3.0-4.9 mm flower buds cultured on the medium supplemented with 1 mg.L^(-1) BA + 1 mg.L^(-1) NAA or 4 mg.L^(-1) BA + 1 mg.L^(-1) NAA. This recipe can be used as initial culture for gene transfer and mutagenic breeding. |
