生甘草對關木通誘發大鼠急性腎毒性之效應

The Effects of Gancao on Guamutong-induced Acute Renal Toxicity in Rats

本研究之目的在探討生甘草對關木通（含馬兜鈴酸）所導致的急性腎毒性的保護作用，研究採用口服餵食給藥之模式，選取48隻Sprange-Dawley（SD）雌性大白鼠，隨機分成：A空白對照組、B1關木通（10g/kg）組、B2關木通（10g/kg）加生甘草（1g/kg）組、C1關木通（20g/kg）組、C2關木通（20g/kg）加生甘草（1g/kg）組、D1關木通（30g/kg）組、D2關木通（30g/kg）加生甘草（1g生藥/kg）組、E生甘草（1g/kg）組，每組6隻大鼠，共計8組。經過1週的餵食之後，記錄其實驗前後之體重、活動力的變化以及其腎臟組織病理變化；檢驗其腎功能血中Blood Urea Nitrogen（BUN）及Creatinine（Cr）的數值。分別以不同劑量的關木通組互相比較，另外再將關木通組分別與同劑量之關木通配伍生甘草組之結果以Fisher’s exacttest及Kruskal-wallis test進行統計分析，並以Duncan’sMultiple-Range Test作事後檢定。研究結果顯示隨著給予關木通之劑量增加，血中之BUN、Cr的數值及腎臟病理病變程度皆隨之上升，活動力及體重增加量則隨之降低。而另添加生甘草後可減緩血中BUN、Cr的數值及腎臟病理病變程度之上升之趨勢。此結果說明使用生甘草配伍關木通，可減輕關木通所造成之急性腎臟傷害，但其機轉為何則仍需進一步探討。

The purpose of this experiment was to investigate how the effect of Gancao on Guamutong (contain Aristolochic acid) - induced acute renal toxicity in rats. Forty-eight female SD rats were randomly separated into eight groups - a control group; a group of 30 g/kg of Guamutong in rat; a group of 30 g/kg of Guamutong combined with 1 g/kg of Gancao in rat; a group of 20 g/kg of Guamutong in rat; a group of 20 g/kg of Guamutong combined with 1 g/kg of Gancao in rat; a group of 10 g/kg of Guamutong in rat; a group of 10 g/kg of Guamutong combined with 1 g/kg of Gancao in rat; a group of 1 g/kg of Gancao in rat. Rats in the eight groups were given different doses of herbs for one week using the gastric tube. We surveyed the general condition, the histology of the kidney, the data of the renal function of blood in the rats. Then we analyzed the results using the SAS packages. Among the groups with different doses of the Guamutong, we found the rats' weight and activity dropped significantly and necrosis area of their epithelium in the renal tubules as well as the biochemical level of BUN and creatinine rose significantly, and these results were dose-dependent. However, these effects of Guamutong were attenuated as feeding the rats with the Gancao. These results indicated that the Gancao may decrease the acute renal toxicity of Guamotong based on the histological and laboratory observations.
