將76株分離自動物及其產製品之金黃色葡萄球菌(生乳62株、鯨魚3株、海豚4株、乳房炎牛乳汁2株、豬2株,為、鼠、犬各1株),以商品化之ELISA套組檢測其腸毒素產生能力,結果有26株產生A型腸毒素,產生B、C型腸毒素者分別有2株及1株,而未檢出產生D、E型腸毒素之菌株。另以自行設計之引子對(primers),以毛細管進行聚合?鏈反應(polymerase chain reaction,PCR)檢測帶有產生A型腸毒素基因之菌株,結果亦有26株呈陽性反應,與ELISA之檢測結果相同,但PCR檢測A型腸毒素基因之靈敏度為104CFU/ml。
A commercial ELISA kit (RIDASCREEN(R) SET A, B, C, D, E. R-Biopharm GmbH, Darmstadt, Germany) was used to examine the enterotoxigenecity of 76 Staphylococcus aureus strains. Of these strains, 62 were isolated from raw milk, 3 from whale, 4 from dolphin, 2 from cow milk with mastitis, 2 from pig, and 1 each from chicken, mouse, and dog. Type A, B, and C enterotoxins were detected in 26, 2, and 1 strains, respectively. Neither type D nor type E enterotoxin- producing strain was observed. In addition, DNA amplification by capillary polymerase chain reaction ( PCR ) with specific primers was used to detect the presence of enterotoxin A gene. The results showed that 26 strains harbored this gene. Study on the detection sensitivity of PCR indicated that the lowest cell number required for positive reaction was 104 CFU/ml." |
