本研究利用高效液相層析儀,分析天麻藥材中之高極性成分parishin、parishin B及C之含量,採用C18逆相層析管柱,移動相為甲醇--0.1%磷酸溶液,利用線性梯度沖提,檢測波長為222 nm;得到良好分析結果。Parishin、parishin B及C三成分之線性迴歸方程式及相關係數(r)分別為:parishin (濃度80~1,200μg/ml), Y= 433.82X-13.90 (r=0.9998),parishin B(濃度120~760μg/ml), Y= 359.03X+55.98 (r=0.9831)及parishin C(濃度40~320μg/ml), Y= 548.45X-60.66 (r=0.9995),均呈現良好線性關f系。添加回收率試驗結果,parishin為102.6 ± 3.3%,parishin B為102.8 ± 2.7%,parishin C為102.6 ± 2.9%,顯示準確性佳。三種指標成分之同日間及異日間試驗之相對標準偏差,分別為0.73~2.11%及0.67~3.56%,顯示高效液相層析法用於天麻指標成分之分析其結果良好。市售天麻藥材23種,利用上述高效液相層析法,定量分析各指標成分parishin、parishin B及C的含量百分比,結果parishin之含量則在0.37~1.50%之間,平均為0.96%;parishin B之含量在0.12~0.65%之間,平均為0.42%;parishin C之含量在0.08~0.27%之間,平均為0.16%。" |

A rapid method by high performance liquid chromatography was developed for the simultaneous determination of three marker substances, parishin, parishins B and C, in Gastrodiae Rhizoma . This determination was carried out by using a gradient elution with methanol and 0.1% phosphoric acid on a Cosmosil 5C18-AR column. The linear calibration range of parishin, parishins Band C were 80~1200µg/ml (r=0.9998). 120~760µg/ml (r=0.9831) and 40~320µg/ml(r=0.9995), respectively. The recoveries of the markers were parishin: 102.6 ± 3.3%, parishin B: 102.8 ± 2.7% and parishin C: 102.6 ± 2.9%. The relative standard deviations of the three marker substances for intraday and interday analyses were 0.73~2.11% and 0.67~3.56%, respectively. The contents of the markers mentioned above in 23 crude drugs of Gastrodiae Rhizoma have been determined and the results were obtained as follows : parishin 0.37~1.50%, parishin B 0.12~0.65% and parishin C 0.08~0.27%. Therefore, HPLC is a useful technique to determine these selected markers. |
