The purpose of this study was to investigate the preventive and therapeutic effect of Gui-Pi-Tang (GPT) on the impairment of water maze performance and hyperglycemia induced by streptozotocin (STZ). We measured oral glucose tolerance and behavior in order to understand the mechanism of the preventive and attenuating effects of GPT, and then provided TCM clinical drug reference. We grouped rats into four categories: the first group: treated with vehicle, the second group: induced by STZ, the third group: STZ-induced diabetic rats pretreated with GPT, the forth group: STZ-induced diabetic rats treated with GPT. The results show, GPT prevention group for oral glucose, blood sugar and blood insulin concentration changes compared with the STZ-induced group, no significant differences; and GPT treatment group for changes in blood glucose after oral glucose lower than the STZ-induced group; and there is no difference in blood insulin concentrations. Secondly, GPT on STZ-induced hyperglycemia study of learning and memory in rats, GPT treatment group significantly improve STZ-induced hyperglycemia in rat spatial learning (p <0.05); GPT treatment or prevention group can improve STZ-induced hyperglycemia rats reference of memory impairment (p <0.05, p <0.01); prevention group STZ-induced hyperglycemia can improve working memory in rats (p <0.01); and treated STZinduced hyperglycemia can improve working memory in rats (p <0.01). From these above results, we found that that GPT given before STZ treatment attenuated STZ-induced diabetic and the deficits in water maze performance, reference memory and working memory. We further suggest that therapeutic effects of GPT in STZ-induced diabetic rats might be due to improving glucose regulation. |
