市售酵素免疫分析(ELISA)套組檢測黃麴毒素之探討

DETECTION OF AFLATOXIN BY COMMERCIAL ELISA KITS

由於傳統定量穀物中黃麴毒素（以下簡稱AF）之高效液相層法（以下簡稱HPLC法），樣品前處理步驟繁複，且需使用大量溶劑，不但不經濟且對操作人員之健康亦可能有所危害，因此發展專一性、敏感性、正確性均高且操作簡單、安全之檢驗方法一直是研究人員努力的方向，目前利用酵素免疫分析法（以下簡稱ELISA法）似可滿足上述需求，且已有許多不同品牌之檢測套組（Kit）上市：本試驗即以市售三種品牌套組，其商品各分別為Agrichek, Chemafla 及 Biotech（詳如表一）檢測己添加5-100 PPb AFB1 之花生，並與衛生署公告之AF檢驗方法（即HPLC法）比較靈敏度，再現性及檢驗成本等以評估市售ELISA檢測套組應用於例行性檢驗之可行性。

In order to investigate the effectiveness of commerical ELISA kits in detecting Aflatoxin, three were evaluated. The results showed that commercial ELISA kits, Used for estimating the content of AF, are generally more sensitive, rapid and cost-effective, compared to the current method. Howerer, these kits could not determine the king and the level of each AF seperately, so, they are very useful for screening and semiquantitatvie testing only.
