The protein constituents of spinach leaf were first investigated by Wildman and Bonner (1947). Two separate classes were istinguished by the different electrophoretic mobilities；these were designated as "Fraction I" and "Fraction II". ln tobacco, spinach and other green leaves, Fraction I protein makes up a major portion of the total leaf protein. Several studies have shown that this rotein has some distinctive properties in addition to its abundance. It has a large molecular weight of approximately 500,000 and exhibits ribulose-1,5-diphosphate carboxylase activity.
