The protein constituents of spinach leaf were first investigated by Wildman and Bonner (1947). Two separate classes were istinguished by the different electrophoretic mobilities;these were designated as "Fraction I" and "Fraction II". ln tobacco, spinach and other green leaves, Fraction I protein makes up a major portion of the total leaf protein. Several studies have shown that this rotein has some distinctive properties in addition to its abundance. It has a large molecular weight of approximately 500,000 and exhibits ribulose-1,5-diphosphate carboxylase activity.
