應用高效液相層析儀同時分析漢防己藥材中指標成分tetrandrine, fangchinoline, cyclanoline 及oblongine 之含量，採用C18 逆向層析管柱，移動相為乙- 磷酸二氫鹽緩衝溶液，利用線性梯度沖提，檢測波長為280 nm ；以上述四種成分作為外插標準；在45 分鐘內四種成分可完全分離，其定量線性回歸方程式及相關係數之濃度在12.5 ～1637 ig/ml 範圍間，檢測極限依S/N=3 規例計算，四種標準品tetrandrine, fangchinoline, cyclanoline 及oblongine 分別為0.95, 0.95, 0.95 及1.69 ig/ml 。A reversed-phase high-performance liquid chromatographic method to simultaneously measure the four alkaloids [tetrandrine, fangchinoline, cyclanoline and oblongine] in Radix Stephaniae tetrandrae has been successfully developed. The method uses standards of the four compounds as external standards. These compounds were isolated in our laboratory using various chromatographic methods and are completely separated within 45 min using a reversed-phase column and linear gradient elution with dihydrogenphosphate buffer HPLC-grade acetonitrile mobile phase. The quantitative calibration curves are linear over a range of 12.5-1637 ig/ml for all four compounds. The detection limits (S/N=3) for tetrandrine, fangchinoline, cyclanoline and oblongine are approximately 0.95, 0.95, 0.95 and 1.69ig/ml, respectively.
