川芎與鬱金所含川芎嗪之高效液相層析定量分析

Determination of Tetramethylpyrazine in the Herbs of Chuanxiong and Yujin by High-Performance Liquid Chromatography

本研究應用一套靈敏、可靠的高效液相層析儀，以逆相碳18的分離管柱（4.6×250mm，粒徑5μm），流速每分鐘1mL來進行中藥材川芎與鬱金之川芎嗪含量分析，移動相為methano1:50 mM NaH2PO4（40:60，v/v，pH3.3以正磷酸調整），並以紫外光檢測器280nm來偵測。取川芎嗪濃度0.1-5μg/mL製成校正曲線。川芎嗪偵測極限為10ng/mL。並由實驗結果得知，於川芎(Ligusticum chuanxiong HORT)沒有偵測到川芎嗪含量，鬱金(Curcuma aromatica SALISB)之正己烷(n-hexane)萃取液所含川芎嗪量為1.66±0.02μg/g。

We used a rapid and reliable high-performance liquid chromatographic method for determination of tetraimethylpyrazine in the medicinal herbs of chuanxiong and yujin. the separation was achieved on a Cosmosil 5C18 column (4.6×250 mm, particle size 5μm). the mobile phase consisted of methanol: 50 mM NaH?烈O??(40:60, v/v, pH 3.3 adjusted by orthophosphoric acid) at a flow rate of 1.0 mL/min. Tetramethylpyrazine was monitored at a wavelength of 280nm. The linear calibration range and detection limit for tetramethylpyrazine was 0.1-5 μg/mL and 10 ng/mL, respectively, It was found that the tetramethylpyrazine was not detected in the Liygusticum chuanxiong HORT. The tetramethylpyrazine contained in n-hexane extract of Curcuma aromatica SALISB was 1.66±0.02 μg/g.
