本文報告對一例因膝部骨關節炎切除的關節軟骨所作的成年人關節軟骨細胞的分離與培養。應用trypsin-collagenase消化法分離細胞。用加有10%胎牛血清、glutamine與gentamicin的F12培養液進行培養。軟骨細胞在體外生長良好,在10個月內傳代15次,細胞數增加為26,562倍,總分裂次數達15次。免疫細胞化學研究(抗S-100抗體)指示這些細胞培養為純軟骨細胞培養,本項研究顯示可用本文敘述之方法建立人類關節軟骨細胞培養。軟骨細胞培養可用於在體外研究關節軟骨細胞之生理及病理,並可用於自體軟骨細胞培養以治療關節軟骨缺損。 |
英文摘要 |
Adult human articular chondrocytes were isolated from surgically excised auricular cartilage from a patient suffering from osteoarthritis of the knee. Cells were isolated by the trypsin-collagenase digestion method and cultured with F12 medium supplemented with 10% fetal bovine serum, glutamine and gentamicin. Cultured chondrocytes grew well. They were able to be passaged for 15 generations and revealed a 26,562-fold increase of cell numbers over a 10 month period (cumulative population doublings=15 times). An immunocytochemical study using anti-S-100 antibodies demonstrated that the culture was a pure cell culture of chondrocytes. The present study indicates that the cell cultures of adult human articular chondrocytes can be established by these methods. Cultured chondrocytes provide an in vitro model system for studying the physiology and pathology of human articular chondrocytes and may be used for autologous transplantation of chondrocytes to treat articular cartilage defects. |
